What are Clinical Trials?

MTT Assay
The MTT assay and test the MTS colorimetric test for measuring the activity of enzymes that reduce or close the dye MTT (XTT, MTS, WSTs) to formazan dye, giving the color purple. MTT (3 – (4,5-Dimethylthiazol-2-yl)-diphenyltetrazolium bromide -2.5, a tetrazole yellow), is reduced to purple formazan in living cells [1] A solubilization solution (usually either dimethyl sulfoxide, a. Ethanol acidified solution, or solutions of sodium dodecyl sulfate detergent in dilute hydrochloric acid) was added to dissolve the insoluble purple formazan product into colored solution. The absorbance of this colored solution can be measured by measuring the specific wavelength (usually between 500 and 600 nm) with a spectrophotometer. The maximum absorption depends on the solvent used.

Finally, tests WST (1) can be read directly (unlike MTT requiring solubilization step), (2) provide a more effective signal than MTT, and (3) reduction of toxicity in cells (such as cell-permeable MTT, and unresolved formazan which accumulates in the cell) [4].

This reduction occurs only when the active reductase enzyme, and therefore conversion is often used as a measure of viable (living) cells.
